Achondroplasia and Chondrodysplasias as Growth-Plate Switch Suppression
Achondroplasia maps FGFR3 gain-of-function to a SUPPRESSIVE drive on the SOX9 chondrocyte switch at the growth plate (§5). FGFR3 normally brakes chondrocyte proliferation; the activating G380R mutation turns that brake up, shifting the growth-plate switch toward OFF and shortening long-bone output. The model gives a graded ladder — full output 1.00 in WT collapsing to 0.09 in achondroplasia — with a clear cliff [V].
Adding a negative regulator to T4: a suppressive drive (FGFR3-GOF) on the chondrocyte switch lowers growth-plate output. The ladder runs WT 1.00 → hypochondroplasia (mild) → achondroplasia 0.09 → thanatophoric (severe), a graded shortening with a threshold — FGFR3 dose maps to severity. A growth-plate brake turned up
T4 ranked developmental thresholds by master-gene γ. Achondroplasia and the related chondrodysplasias perturb that growth plate with a NEGATIVE regulator. FGFR3 is a physiological brake on chondrocyte proliferation; the activating G380R substitution (and its stronger alleles) increases that braking as a sustained SUPPRESSIVE drive on the SOX9 chondrocyte switch, pushing it toward the OFF basin and cutting endochondral long-bone output. It is the inverse perturbation to a master-gene loss.

The output ladder is graded and shows a threshold: a mild suppressive drive (hypochondroplasia) barely dents output, the achondroplasia allele drops it sharply to 0.09, and the severe (thanatophoric) drive collapses it further. Stronger FGFR3 activation → shorter bone, matching the clinical allelic series. The DIRECTION and the graded cliff are the result.
What is NOT claimed
The growth-plate output is a switch-state proxy, so the relative ladder is claimed but the ABSOLUTE long-bone length in centimetres is NOT fixed by the substrate and stays [O].
